Novel Single-Molecule Assay Development for
Neurotransmitter Exocytosis

1. Content &Characteristic

O This project develops new assay to study the roles of various SNARE proteins, accessory proteins and the effects of different lipid compositions for fusion steps in neurotransmitter exocytosis.

- We developed a single-molecule FRET-based vesicle-vesicle fusion assay for real time observation of lipid-mixing and content-mixing incorporating SNARE proteins.

- By using our new single-molecule assay, we detect fusion pore expansion and propose the roles of SNARE protein in SNARE-dependent membrane fusion.



2. Scientific Value

O Real-time observation at the single molecule level of neurotransmitter exocytosis can unveil a detailed description of its dynamics and further more introduce a new model.

O Incorporating dementia inducing proteins to our assay, the expected outcomes will establish a new concept of the pathochemical origin of dementia.

Economic Effect

O Once proven the outcomes will revolutionize our understanding of dementia which will lead to development of new drugs, drug testing platforms, prevention and early diagnosis for the treatment of dementia.
